Useful Cell Line (PMDC05) of Plasmacytoid Dendritic Cell for Custom made Immunotherapy
We established a unique leukemic pDC cell line, PMDC05 cells, which grow in the absence of cytokines as well as feeder cells. There are two cell lines reported to be derived from leukemia cells of the diseases in the pDCL category. One is the pDC lines, GEN2.2, which was developed from a patient with pDCL and cultured on MS-5 feeder cells, and the other is a pDC line, CAL-1, which was established from a patient with blastic natural killer cell lymphoma.
Similar to normal pDC, PMDC05 possess a potent antigen presenting ability to naive T cells and can produce IFN-a by stimulation with CpG and influenza virus. On the contrary, by stimulation with LPS, PMDC05 can produce IL-12p70. A further study showed a shift of pDC-like cells to mDC-like cells in PMDC05.
The pDC cell line, PMDC05, could be a useful tool for investigating the pathophysiology of pDCL and exploring the nature of normal pDCs.
